WPAD auto detect vs use automatic Script?

Hi, I was wondering which would be faster when implementing WPAD, As right now all users use auto detect, but sometimes chrome takes a while to find the proxy, would it be better to uncheck auto detect and use the configuration script or can i have both checked? if so would chrome first look for the auto detect or the config script?
