Can you run a PPPoE connection over an interface which already has an IP-configuration, lets say dhcp?

Yes. Many people who run PPPoE have an additional IP configuration on the same network port to access the modem.

Go to Interfaces > Assignments > PPPs, hit Add, select PPPoE from link type drop-down and the respective interface.
Enter your credentials and a description below and save the settings.
The PPPoE connection should be established immediately if the settings are correct.

You wouldn't normally use PPPoE on an interface configured as DHCP but you could.

What exactly are you trying to do?

@stephenw10 There is a router that also allows PPPoE Passthrough and I want to use only one connection.

Putting a device in passthrough mode like that usually disables DHCP but if it doesn't I see no reason why it wouldn't work. I have configured static IPs on PPPoE parent NICs to access the modem many times. I have that setup here.
