I need to setup this lab and I am curious if it can be done with pfsense. Can anyone help me out?

Basically I need a DMZ to the VM box and all lans need to be behind a NAT. The mgmt network is not routed.

Absolutely, but you will either need 5 NIC's or a switch that can do L3 and/or vlan tagging.
